Bovine herpesvirus type 1 in cumulus-oocyte complexes collected from naturally infected cows PAB

Abstract: The objective of this work was to investigate the presence of bovine herpesvirus type 1 (BoHV-1) in follicular fluid and in cumulus-oocyte complexes (COC) recovered from naturally infected cows but with no clinical signs of the disease. Cows that were seropositive (n=38) or seronegative (n=8, control) to infectious bovine rhinotracheitis were selected after a serum neutralization test in microplates. The presence of the virus was investigated by PCR in COC and in follicular fluid. Viral DNA was not found in any of the samples. The obtained results suggest that serologically positive cows with no clinical signs of the disease offer negligible risk of transmitting BoHV-1 by COC or follicular fluid.

Detection of virulence genes and the phylogenetic groups of Escherichia coli isolated from dogs in Brazil Ciência Rural

ABSTRACT: This study identified the virulence genes, pathovars, and phylogenetic groups of Escherichia coli strains obtained from the feces of dogs with and without diarrhea. Virulence genes and phylogenetic group identification were studied using polymerase chain reaction. Thirty-seven E. coli isolates were positive for at least one virulence factor gene. Twenty-one (57.8%) of the positive isolates were isolated from diarrheal feces and sixteen (43.2%) were from the feces of non-diarrheic dogs. Enteropathogenic E. coli (EPEC) were the most frequently (62.2%) detected pathovar in dog feces and were mainly from phylogroup B1 and E. Development and validation of a modified TaqMan based real-time PCR assay targeting the lipl32 gene for detection of pathogenic Leptospira in canine urine samples BJM

Abstract A modified TaqMan real-time polymerase chain reaction targeting a 138 bp fragment within the lipl32 gene was developed to identify exclusively pathogenic Leptospira spp. in dog urine samples. Thirty-five samples from dogs with suspected clinical leptospirosis and 116 samples from apparently healthy dogs were tested for presence of leptospiral DNA using the TaqMan-based assay. The results were compared with those from a well-established conventional PCR targeting the 16S RNA encoding gene associated with nucleotide sequencing analysis. ABSTRACT: Pseudorabies (PR) is a highly contagious viral disease of great animal health and economic importance in swine industry. The aim of this study was to evaluate different genomic regions, real-time PCR chemistries and equipment for the molecular diagnosis of PR. Eight primer pairs targeting four genes (gB, gC, gE, gD), three different qPCR chemistries (SybrGreen, hydrolysis probes and plexor) and two equipment (ABI7500, Rotorgene 3000) were evaluated. O conhecimento sobre a fisiologia e regulação dos eletrólitos e do equilíbrio ácido-base é necessário, pois a saúde do animal depende diretamente da composição normal dos fluídos nos diversos compartimentos corporais. Infectious abortion is a significant cause of reproductive failure and economic losses in cattle. The goal of this study was to detect nucleic acids of several infectious agents known to cause abortion including Arcanobacterium pyogenes, Bovine Herpesvirus 1, Brucella abortus, Campylobacter fetus subsp. venerealis, Chlamydophila abortus, Leptospira sp., Listeria monocytogenes, Salmonella sp., Mycoplasma bovis, Mycoplasma bovigenitalium, Neospora caninum, and Tritrichomonas foetus. Frequency and antimicrobial susceptibility of Staphylococcus pseudintermedius in dogs with otitis externa Ciência Rural

ABSTRACT: Infections by Staphyloccocus pseudintermedius in the skin of dogs have been object of studies, since such microorganisms often present multiple resistance to antibiotics. This study aimed to identify and evaluate the antimicrobial susceptibility of Staphylococcus pseudintermedius (SP) strains isolated from dogs with otitis. Swabs from both ears of 52 dogs with otitis (n=104) were included. Bacteria were cultured using Muller-Hinton agar (supplemented with 5% equine blood and incubated at 37°C for 24 to 48 hours. All colonies underwent biochemical evaluation for identification of staphylococci. The identity of colonies as SP was confirmed by polymerase chain reaction. Isolation and identification of Mycobacterium bovis in milk from cows in northeastern Brazil Ciência Rural

ABSTRACT: Milk samples from 16 cows that tested positive on the tuberculin test in the state of Paraíba, northeastern Brazil, were used for mycobacteria isolation and identification. Mycobacteria were isolated from five (31.25%) of the 16 milk samples; three samples were classified as M. bovis, and two as belonging to the Mycobacterium genus. This is probably the first study of isolation and identification of M. bovis in milk from cows in Northeastern Brazil, which suggests that humans are at risk of contamination by ingestion.

Seroprevalence of antibodies against group A rotavirus in cattle from a pioneer frontier in Brazilian Amazon Braz. J. Vet. Res. Anim. Sci.

Rotavirus is a worldwide etiologic agent of diarrhea, responsible for large economic losses. We studied the seroprevalence of antibodies to group A rotavirus in cattle in 67 smallholder farms from Uruará municipality, using counterimmunoelectroosmophoresis with the NCDV strain as a standard antigen. Prevalence of positive smallholder farms was 95.6-100%. Significant differences were seen between age groups when the seropositivity rose from the youngest to the oldest groups and between females and males older than 1 year, when the seropositivity was higher in the first group.

Validation of a real-time PCR assay for the molecular identification of Mycobacterium tuberculosis BJM

Mycobacterium tuberculosis is the major cause of tuberculosis in humans. This bacillus gained prominence with the occurrence of HIV, presenting itself as an important opportunistic infection associated with acquired immunodeficiency syndrome (AIDS). The current study aimed to develop a real-time PCR using Eva Green technology for molecular identification of M. tuberculosis isolates. The primers were designed to Rv1510 gene. Ninety nine samples of M. tuberculosis and sixty samples of M. bovis were tested and no sample of the bovine bacillus was detected by the qPCR. Virulence factors and phylotyping of Escherichia coli isolated from non-diarrheic and diarrheic water buffalo calves Ciência Rural

ABSTRACT: This study aimed to determine the virulence factors, phylogenetic groups, and the relationships between pathovars and phylogenetic groups of E. coli strains isolated from feces of buffalo calves. A total of 217 E. coli strains were obtained from feces after culture and were screened by PCR for detection of virulence factors EAST-1, enterohemolysin, Saa, CNF2, F41, F5, STa, intimin, Stx1 and Stx2. One hundred and thirty-four isolates were positive for one or more virulence factors: eighty-four from diarrheic animals, and fifty from non-diarrheic calves.
